of a new technical poster demonstrating the use of its purpose-built Thermo
Scientific Galileo LIMS™ to perform in vitro ADME/Tox profiling. The new
technical poster is available free-of-charge via www.thermo.com/galileo. Thermo
Scientific Galileo Laboratory Information Management System (LIMS) provides an
integrated, template-driven database system to allow rapid data review and
acceptance for large numbers of compounds for a range of ADME/Tox profiling and
screening applications such as metabolic stability, IC50, Caco-2 and PAMPA. The
solution enables pharmaceutical and CRO companies to manage data efficiently and
appropriately in order to rapidly execute key decisions on drug candidate
selection.
ADME/Tox profiling has become increasingly important for
efficient drug candidate selection and optimization of a compound's
physicochemical properties. Point solutions such as standalone spreadsheet and
graphing tools are inefficient, prone to errors and lead to inconsistent
results. Template modifications propagated over time cause variations in data
handling and problematic 'apples to apples' comparisons. The lack of a central
repository results in data being maintained in disparate locations without
traceability. A fully functional database system can reduce effort and manpower
required and increase throughput by automating complex experiment templating,
instrument interfacing, data analysis as well as review and acceptance
tasks.
Thermo Scientific Galileo LIMS provides simple,
easy-to-use data processing, graphing, data review and reporting while also
facilitating consolidation and automation of multiple processes. Furthermore,
flexible calculation settings enable laboratories to follow their standard
procedures. Experimental set-up is fast and easy requiring no programming.
Moreover, the system is capable of performing automatic re-calculation after
outlier data is removal. User-definable calculations using Microsoft Excel
spreadsheets are controlled and stored securely in Galileo LIMS database, adding
security and allowing GLP compliance if necessary. Users are allowed to set up
their own tree-style logic for flagging of results.
The study design of Thermo Scientific Galileo LIMS
templates allows specification of key experimental variables including drug and
inhibitor concentrations, species, biomatrix, tissue, analytical time points,
assay methods and result calculation options. Manual data transfer to external
software for processing and storage is virtually eliminated. Instrument data can
be rapidly imported via Rapid Study Entry using a simple dialog to populate the
template with analytical results and hence automatic calculation of ADME/Tox
parameters.
The system's Gallery presents data in tabular, thumbnail
or detailed views. The thumbnail view allows users to scan quickly across many
datasets while the details view allows users to 'drill-down' into a data set.
Results are automatically calculated when the charting galleries are opened and
re-calculated whenever outlier points are deactivated. ADME/Tox parameters can
be flagged automatically in the Gallery or in the Details view using green,
yellow or red flags to indicate acceptable, questionable or rejected results for
each drug candidate.
